Personally, I'd pick it off with my fingers (grounding myself first, to one extent or another, depending on the humidity), then I'd clean it with isopropyl alcohol and a cue-tip.

Serial read and write speeds aren't that different between 5400 rpm and 7200 rpm drives since you get get higher densities with 5400rpm drives. So backup times shouldn't be that different. Where it makes a difference is in random access speeds
