Very easy, as long as you have a proper drill and bit.
Get a drill bit that's slightly smaller than the hole so that you don't damage the case. I did this to an old Antec SX1030 and was able to use standard case screws in place of the rivets.

Yes. It's also easy to put them back in. Most computer rivets are 1/8" so all you need is a drill and a 1/8" bit. I had more of an issue getting the spare parts of the rivet off of the bit than drilling anything out (pliers or vise-grips are helpful.)
If you don't want/can't replace them with screws most hardware stores will have a riveter for around $20.
