No enclosure that I'm aware of will silence a noisy hard drive.

For most of these enclosures, you'd need a quiet drive to start off with.

My system is inaudible when idle and I bought a Maxtor Maxline Pro 500GB hard drive and put it inside a Vantec eSATA enclosure. When I turn the external drive on, it becomes the loudest thing on my desktop. The reason I chose external is I only turn it on 2-3 times a day and I know if it was internal it would make my system audible (I usually have it on 24-7 in my bedroom and I need it completely silent when I go to sleep).

Other things to consider: heat and speed. Most enclosures don't come with a fan, so the drive can overheat quite easily. My external 500GB can shoot up to 55C if I leave it on a couple of hours. Also if you're using USB, it will be much slower than internal. If you're using Firewire it's somewhat faster (than USB), but if you're using eSATA it's as fast as an internal drive.
